The article discusses families near Medellín, Colombia who carry a rare genetic mutation, called the Paisa mutation, that causes Alzheimer's disease. Topics include Alejandra, whose mother developed early-onset Alzheimer's when she was 16 years old, familial Alzheimer's in Antioquians, neurologists Francisco Lopera and Kenneth S. Kosik's discovery of the Paisa mutation in 1995, and a clinical trial of the drug crenezumab in Medellín, called the Alzheimer's Preventive Initiative.
