The Wnt signaling pathway in bone metabolism: Biology and therapeutic interventions. (English)

Wnt signaling has been shown to be an important regulatory pathway in the osteogenetic differentiation of mesenchymal stem cells. Induction of the Wnt signaling pathway promotes bone formation, bone renewal and differentiation of osteoblasts and osteoclasts, while inactivation of the Wnt pathway leads to osteopenia. Activation and inactivation of Wnt signaling deviations during osteogenesis lead to sclerosis and osteoporosis, respectively. Study of the Wnt signaling mechanism and its interactions with other signaling pathways in bones provides potential therapeutic targets for the treatment of bone diseases. Although modification of the Wnt signal pathway has a variety of therapeutic possibilities, careful handling is required, because of the risk of oncogenesis. This paper reports on the role of Wnt signaling in bone metabolism, the potential therapeutic targets, and the risks arising from intervention in Wnt signaling. [ABSTRACT FROM AUTHOR]

Η σηματοδότηση Wnt έχει δειχθεί ως ένα σημαντικό ρυθμιστικό μονοπάτι στην οστεογενετική διαφοροποίηση των μεσεγχυματικών βλαστικών κυττάρων. Επαγωγή του μονοπατιού σηματοδότησης Wnt προάγει τον σχηματισμό των οστών, την οστική ανανέωση, καθώς και τη διαφοροποίηση των οστεοβλαστών και των οστεοκλαστών, ενώ η αδρανοποίηση της οδού οδηγεί σε οστεοπενία. Επίσης, ενεργοποίηση και αδρανοποίηση των εκτροπών της κανονικής οδού σηματοδότησης Wnt στην οστεογένεση οδηγεί στη σκληροστέωση και στην οστεοπόρωση, αντίστοιχα. Η μελέτη του μηχανισμού σηματοδότησης Wnt και των αλληλεπιδράσεών του με άλλα μονοπάτια σηματοδότησης στα οστά παρέχει πιθανούς θεραπευτικούς στόχους για τη θεραπεία των εν λόγω οστικών παθήσεων. Μολονότι η τροποποίηση του σηματοδοτικού μονοπατιού Wnt έχει πληθώρα θεραπευτικών δυνατοτήτων, απαιτούνται προσεκτικοί χειρισμοί λόγω του κινδύνου της ογκογένεσης. Στην παρούσα εργασία παρουσιάζεται ο ρόλος της σηματοδότησης Wnt στον οστικό μεταβολισμό, οι δυνητικοί θεραπευτικοί στόχοι, αλλά και οι κίνδυνοι που προκύπτουν από την παρέμβαση στη σηματοδότηση του Wnt.
