      Supplemental Figure 5. Activity of PAV-951-treated BB1 and BB5 cells. BB1 (benign adenoma) and BB5 (malignant adenocarcinoma) cell lines were derived from two patient biopsy samples. Cells were treated with compound and stained for Hoechst (Blue) and Chromalive (Yellow and Red) then imaged by spinning disk confocal microscopy with a 20X water immersion lens (Opera Phenix). Supplemental Figure 5A and 5B show dose titrations of PAV-617 and PAV-951 respectively on BB1 and BB5, where IC50 is calculated as the percentage of cells (identified by segmentation based on Hoescht staining) that showed loss of mitochondrial activity (loss of yellow staining). Supplemental Figure C, primary cell cultures treated with DMSO (vehicle). Upper panels show staining with Hoescht and Chromalive yellow (indicates mitochondrial activity), lower panels show staining with Hoescht (blue), Chromalive (yellow, mitochondrial activity) and (red, indicates most non-mitochondrial cellular membranes). Supplemental Figure D, Upper panels BB1, Lower panels BB5, primary cells treated with the concentrations of PAV-951 indicated above the panels. Cells were stained with Hoescht (blue) and Chromalive (yellow and red). This experiment was conducted once, with four replicates per condition.
